The Hakodate Main Line is a railway line that connects Hakodate to Asahikawa via Oshamanbe, Otaru, and Sapporo. The line from Asahikawa to Otaru is double-tracked and electrified with AC 20kv, 50hz. On this section, the limited express train "Lilac" and "Kamuy" run twice every hour from Asahikawa to Sapporo, and the limited express "Soya" runs once a day from Sapporo to Wakkanai on the Soya Main Line via Asahikawa, and "Okhotsk'' runs from Sapporo to Abashiri on the Sekihoku Main Line via Asahikawa twice a day. In addition, the limited express "Sarobetsu" between Asahikawa and Wakkanai on the Soya Main Line and the "Taisetsu" limited express between Asahikawa and Abashiri on the Sekihoku Main Line connect to the limited express "Lilac" at Asahikawa Station. "Lilac" and "Kamuy" are operated by 789 series AC EMU trains, and "Soya" and "Okhotsk" are operated by diesel trains. The maximum speed of limited express trains is 120km/h. The section from Asahikawa to Fukagawa has a series of tunnels along the Ishikari River. The Ishikari River is the third longest river in Japan after the Shinano River and the Tone River, and the second largest river basin area after the Tone River. The Rumoi Line branches off from Fukagawa Station. The Rumoi Line used to extend from Rumoi to Mashike, towns facing the Sea of Japan, but it was discontinued due to a decrease in ridership. As of 2024, it is a short line of about 15 km, connecting Fukagawa to Ishikari Numata.. The section from Fukagawa to Ishikari Numata is also scheduled to be abolished in 2026.

At Takikawa Station, the Nemuro Main Line (Ashibetsu Subdivision) joins. This line used to connect Takigawa from Nemuro, the easternmost tip of Hokkaido, but after the section from Shintoku Station to Furano Station on the way was damaged by a disaster, it was abolished without being restored, and now the section is closed. This line connects Furano, which is famous for its lavender, and Takikawa. During the season, special express trains run from Furano to Sapporo.
The limited express "Soya" and the limited express "Okhotsk No. 2" bound for Sapporo pass through Sunagawa Station and Bibai Station. All express trains other than these two trains stop.

Most of the trains operated in the section from Asahikawa to Iwamizawa are limited express trains and there are only a few local trains, but from Iwamizawa, many local trains are operated towards Sapporo as it enters the commuter town of Sapporo, the largest city in Hokkaido. Additionally, the Oiwake Subdivision of the Muroran Main Line branches toward Tomakomai. The section of the Muroran Main Line from Iwamizawa to Numanohata (one station before Tomakomai Station) was once a thriving area for coal transportation, and the entire line was double-tracked. Coal transport has declined, and currently only a few local trains and freight trains run from northern and eastern Hokkaido to southern Hokkaido and Honshu. Some sections that were once double track have been replaced with single track after the track was removed in areas affected by disasters.
After passing Atsubetsu Station, the Hakodate Main Line merges with the Chitose Line from New Chitose Airport and Tomakomai, forming a quadruple-track line to Sapporo Station. Only local trains on the Chitose Line stop at Heiwa Station, and local trains on the Hakodate Main Line pass the station.
Sapporo Station is a station with six platforms and 10 rail tracks, and is the last stop for limited express trains. Some local trains run directly to the Hakodate Main Line towards Otaru or the Gakuen Toshi Line. Sapporo Station, which will serve the Hokkaido Shinkansen from Tokyo, will be built in the future on the southeast side of the station.

Two types of limited express trains run on the Hakodate Main Line between Sapporo and Asahikawa: "Lilac'' and "Kamuy.'' "Lilac" uses the 789-0 series, which has a six-car train, and is connected to a half-chamber green car. "Kamuy" uses the 789-1000 series, which is a five-car train, and consists of only regular cars. "Lilac" and "Kamuy" have the same stops and travel time, and both take 1 hour and 25 minutes to cover 136.8 km from Asahikawa to Sapporo.

The 789-0 series used on the limited express “Lilac” was operated as the limited express “Super Hakucho(Swan)” connecting Honshu and Hakodate before the Hokkaido Shinkansen opened from Shin-Aomori Station in Honshu to Shin-Hakodate-Hokuto Station in Hokkaido. After the Hokkaido Shinkansen opened to Shin-Hakodate-Hokuto Station, this trainsets were transferred to “Lilac”, and they replaced the older 785 series train.

The 789-1000 series used in the limited express "Kamuy" are trainsets manufactured to replace the 781 series, which are JNR type trainsets. The train used to run from New Chitose Airport to Asahikawa under the name "Super Kamuy." In addition, the train was operated from New Chitose Airport to Sapporo Station as the rapid train "Airport", and at Sapporo Station the train turned around and was operated as the limited express "Super Kamuy" to Asahikawa.
The 789 series 1000 series is also used for the limited express "Suzuran'' that connects Sapporo to Muroran via Tomakomai.
